Mechanism Action Of Entavir 0.5mg
Entecavir is an antiviral drug. Entecavir, a guanosine nucleoside analogue with activity against HBV reverse transcriptase (rt), is efficiently phosphorylated to the active triphosphate form, which has an intracellular half-life of 15 hours. By competing with the natural substrate, deoxyguanosine triphosphate, entecavir triphosphate functionally inhibits all three activities of the HBV reverse transcriptase: (1) base priming, (2) reverse transcription of the negative strand from the pregenomic messenger RNA, and (3) synthesis of the positive strand of HBV DNA. Entecavir triphosphate is a weak inhibitor of cellular DNA polymerases α, β, and δ and mitochondrial DNA polymerase gamma with Ki values ranging from 18 to >160 μM.

Exacerbation of Hepatitis B – Discontinuation of Entavir 0.5 mg Tablet or any other anti-Hepatitis B therapy may cause severe acute exacerbation of the HBV infection. Monitoring of liver function is recommended for at least a few months after stopping the treatment with this medicine. Further initiation of anti-Hepatitis B therapy may be warranted in a few cases.
HIV co-infection – Entavir 0.5 mg Tablet is not recommended for use in patients who are infected with human immunodeficiency virus (HIV) and not on highly active antiretroviral therapy (HAART) therapy for HIV due to the risk of development of resistance to HIV NRTIs.
Lactic Acidosis – This medicine is not recommended for use in patients with severe liver dysfunction due to the risk of lactic acidosis (increased acid in the blood). Frequent clinical monitoring for lactic acidosis is recommended while taking this medicine. Any symptoms like difficulty in breathing, irregular heartbeats, dark-coloured urine, and yellow colored skin and eyes should be reported to the doctor on priority.
Pregnancy – This medicine is not recommended for use in pregnant women unless strictly indicated. The benefits and risks should be discussed with the doctor before using this medicine.
Breast-feeding – This medicine is not recommended for use in breastfeeding women unless absolutely necessary. Your doctor may advise you to discontinue breastfeeding or to discontinue the medicine based on your clinical condition.
